Mycobacterial culture involves growing a tissue or fluid sample under laboratory conditions to see whether Mycobacterium tuberculosis grows, which confirms active infection with more certainty than PCR alone. It is considered a reference-standard test under NTEP guidance, though it takes considerably longer to give a result (often several weeks) and can still be negative even when TB is present, because the organism can be sparse in genital tissue. In suspected genital tuberculosis, culture is typically used together with histopathology, PCR, and imaging rather than as a single deciding test, given how slow-growing and patchy this form of TB can be.
